Researchers recently evaluated zanidatamab gastroesophageal cancer therapy in a groundbreaking Phase 3 trial for HER2-positive patients. This dual HER2-targeted bispecific antibody shows significant promise as a first-line treatment option. Therefore, the study compared zanidatamab, both with and without the anti-PD-1 agent tislelizumab, against the standard-of-care trastuzumab and chemotherapy regimen.
The trial results demonstrate that zanidatamab-based regimens significantly extend progression-free survival. Specifically, patients receiving zanidatamab plus chemotherapy achieved a median progression-free survival of 12.4 months. Similarly, those receiving the triplet combination including tislelizumab also saw a median of 12.4 months. In contrast, the control group receiving trastuzumab and chemotherapy reached only 8.1 months. Consequently, these findings suggest a superior alternative for HER2-positive advanced adenocarcinoma management.
Safety profiles remained manageable across all treatment arms. Although adverse events occurred, they aligned with the known profiles of these specific agents. Furthermore, the overall survival data trended in favor of the zanidatamab groups. As a result, clinicians should consider these results when determining optimal first-line strategies for metastatic gastroesophageal cancer.
Zanidatamab is a bispecific antibody designed to bind two distinct HER2 epitopes. This mechanism enhances HER2 signal blockade and promotes receptor internalization compared to traditional monoclonal antibodies.
In this Phase 3 trial, zanidatamab-based regimens reduced the risk of disease progression or death by approximately 35% to 37% compared to trastuzumab. Both zanidatamab arms showed a median progression-free survival of 12.4 months versus 8.1 months for trastuzumab.
