Recently, researchers identified a critical spatial regulator of cellular movement. Specifically, the collective cell migration mechanism relies on a precise balance between protrusion and contraction. However, how cells coordinate these opposing forces in vivo has remained unclear until now. Furthermore, a new study published in the Journal of Cell Biology used Drosophila models to investigate these complex interactions.
Consequently, the research team discovered that RhoGAP15B inhibits RhoA at the leading edge of migrating clusters. Moreover, this spatial inhibition prevents excessive actomyosin contractility at the front. In addition, this localized relaxation allows cells to form stable, directed protrusions. Therefore, RhoGAP15B effectively promotes the forward movement of cell clusters. Notably, the study systematically characterized all RhoGEFs and RhoGAPs in border cells. As a result, they pinpointed RhoGAP15B as a key spatial regulator of RhoA. Finally, these findings highlight how patterned activities drive protrusion dynamics during migration.
RhoGAP15B acts as a spatial inhibitor that reduces RhoA activity at the front of a cell cluster. This action lowers contractility, which allows the leading cells to extend protrusions more effectively.
RhoA regulates actomyosin contraction and cell adhesion. Since collective migration is a hallmark of cancer metastasis, understanding regulators like RhoGAP15B may reveal new targets for inhibiting the spread of tumor cells.
