Recent clinical evidence highlights the importance of podoplanin expression in OSCC as a vital indicator of disease progression. Oral Squamous Cell Carcinoma (OSCC) remains a significant global health challenge, particularly in the Indian subcontinent. Because of its aggressive nature, clinicians require reliable biomarkers to predict metastasis and patient outcomes. Consequently, researchers investigated podoplanin, a mucin-type transmembrane glycoprotein, to assess its prognostic utility in a cohort of 110 Indian patients.
The study reveals that nearly 64.5% of OSCC cases demonstrate strong podoplanin expression using the German Immunoreactive Score. Furthermore, this overexpression shows a significant positive correlation with larger tumor dimensions and lymph node metastasis. For instance, tumors exceeding 2.5 cm frequently exhibited higher staining intensities. Additionally, the presence of nodal involvement (N1-N3) strongly associated with elevated podoplanin levels. Interestingly, while the study showed these links, it did not find a direct correlation with the overall pathological stage or survival rates.
Moreover, the researchers observed that the expression was primarily peripheral and membranous. This pattern suggests that podoplanin plays a functional role at the tumor invasion front. Therefore, it likely contributes to early carcinogenic events and local spread. Although the survival analysis did not reach statistical significance in this specific group, a clear trend toward higher mortality appeared in patients with strong expression. Therefore, podoplanin remains a promising diagnostic and prognostic adjunct in oral oncology.
In addition, clinicians should consider podoplanin levels when planning surgical and therapeutic interventions. Because oral cancer diagnosis often occurs at advanced stages, such biomarkers help in risk stratification. While well-differentiated tumors showed strong expression in this study, poorly differentiated cases sometimes showed weaker patterns. Overall, these findings support the growing evidence of podoplanin's role in tumor biology and growth. Further multicentric studies are recommended to validate these observations for broader clinical use.
