Docosahexaenoic acid (DHA) is a critical nutrient for neurodevelopment in infants and pregnant women. However, the potential for contamination by per- and polyfluoroalkyl substances (PFAS), known as "forever chemicals," poses a significant health risk. Clinicians often recommend algal or fish oil products, but the absence of standardized testing for PFAS in DHA supplements has previously hindered effective risk assessment. PFAS are persistent environmental pollutants that bioaccumulate in the food chain. Because these chemicals do not break down easily, they can infiltrate marine-based oils used in supplements. Exposure during pregnancy is particularly concerning because PFAS can cross the placenta. This exposure may lead to adverse outcomes such as low birth weight, immune suppression, and developmental delays. Consequently, the need for reliable quality control in the nutraceutical industry is more urgent than ever.
A recent study developed and validated a sensitive LC-MS/MS method for the simultaneous determination of four major PFAS: PFOA, PFODA, PFOS, and PFBS. The research team tested various DHA matrices, including both algal and fish oils. They utilized a rapid 12-minute chromatographic separation technique that achieves high sensitivity. The method showed excellent recovery rates between 92.1% and 108.4%, ensuring that even trace amounts of contaminants are identifiable.
During the analysis of thirteen commercial DHA products, the researchers detected trace amounts of PFAS in three samples. Although these levels remained below current regulatory limits, the finding underscores the necessity for routine monitoring. Furthermore, the use of matrix-matched calibration effectively corrected potential interference from the oil-based samples, making this method a practical tool for laboratory quality control.
In India, the use of DHA supplements among pregnant women and infants is widespread. Meanwhile, the Food Safety and Standards Authority of India (FSSAI) has recently proposed stricter regulations regarding PFAS in food contact materials. This technological advancement provides Indian clinicians and manufacturers with the technical support needed to ensure that supplements are free from toxic residues. Pediatricians and obstetricians can now look forward to more rigorous safety standards for the products they prescribe.
PFAS exposure during pregnancy can disrupt endocrine function and interfere with placental health. Studies link these chemicals to reduced birth weight and impaired lung and immune system development in children.
The new LC-MS/MS method is applicable to both matrices. While algal oil is often perceived as having a lower contamination risk due to controlled cultivation, trace PFAS can still occur, making testing essential for both types.
Clinicians should continue recommending DHA for its proven benefits but encourage patients to choose reputable brands that utilize advanced quality control methods for contaminant screening.
