Other noteworthy leads include Simalikalactone D and Verrucarin L acetate, both of which display nanomolar potency against various Plasmodium strains. These findings offer a roadmap for developing the next generation of anti-malarial drugs.
Analogue studies have successfully improved the efficacy of these natural molecular scaffolds. Furthermore, scientists now employ advanced isolation techniques to streamline the identification of bioactive metabolites from complex extracts. These multiple approaches are necessary to discover agents that are both potent and safe for human use. By optimizing these lead compounds, researchers aim to overcome the challenges of bioavailability and toxicity that often hinder natural product development.
Emerging resistance to artemisinin-based therapies threatens current global control efforts. Therefore, the discovery of new molecular scaffolds with different mechanisms of action is essential to prevent a resurgence of malaria deaths.
New leads are emerging from diverse environments. These include terrestrial plants such as Quassia amara (the source of Simalikalactone D) and marine organisms like the sponge Plakortis lita (the source of Thiaplakortones).
