Researchers identify that extracellular vesicles (EVs) from aged individuals trigger mitochondrial dysfunction in HPCs. These vesicles serve as vital communication channels between cells throughout the body. However, aging significantly alters the cargo within these nanoparticles. Consequently, this change negatively impacts the health of haematopoietic progenitor cells. This process highlights a critical mechanism of systemic aging.
Specifically, the accumulation of mutated mitochondrial DNA (mtDNA) within aged EVs leads to metabolic failure. When these vesicles enter young progenitor cells, they actively suppress respiratory function. Moreover, this process facilitates a state of chronic low-grade inflammation, often termed \"inflamm-ageing.\" Therefore, the systemic environment directly shapes the aging process of the blood system. Notably, researchers observed that the concentration of vesicles remains stable while their biological quality declines over time.
While young vesicles support cell health, aged vesicles impair mitochondrial respiration. Furthermore, this dysfunction contributes to the development of clonal hematopoiesis. In addition, these changes increase the risk of hematologic malignancies in the elderly. Consequently, targeting these vesicles might offer a new pathway for geriatric medicine and regenerative therapy. Ultimately, this discovery provides a clearer link between systemic aging and age-related blood disorders. Doctors must consider these non-cell-autonomous factors when evaluating hematologic health in older patients.
Aged extracellular vesicles carry mutated mitochondrial DNA and damaged proteins. When haematopoietic progenitor cells absorb this cargo, it impairs their mitochondrial respiration and reduces their overall regenerative capacity.
Mitochondrial dysfunction in these cells reduces their ability to produce healthy blood lineages. This failure often leads to anemia, weakened immunity, and an increased risk of clonal hematopoiesis in older adults.
