The Nev-RA system functions through a spatiotemporally controlled process. First, angiopep-2 peptides facilitate the penetration of the blood-spinal cord barrier. Once the carrier reaches the lesion site, reactive oxygen species (ROS) trigger the cleavage of the protective coating. This action exposes oligoarginines that effectively bind and neutralize cfDNA. Therefore, this prevents toll-like receptor 9 (TLR9) from initiating a damaging inflammatory cascade.
Furthermore, the extracellular vesicles (Nev) provide substantial neuroprotective benefits. They support the survival of impaired neurons and boost the intrinsic repair capacity of the spinal cord tissue. Additionally, the system addresses the dual barriers of a hostile microenvironment and insufficient natural regeneration. This paradigm represents a significant step forward in neurotrauma therapy. Scientists believe this biomimetic strategy could revolutionize how we treat acute spinal injuries in the future.
Cell-free DNA acts as a damage-associated molecular pattern (DAMP). It binds to TLR9 receptors, which induces chronic neuroinflammation and leads to the death of healthy neurons, thereby hindering recovery.
