Recent research identifies a critical pathway involving NINJ2 and Colitis Healing. Activation-induced cytidine deaminase (AID), usually associated with antibody diversity in B cells, also plays a pivotal role in the gut. Scientists found that AID is significantly expressed in colonic epithelial cells (CECs) during active inflammation. This discovery sheds light on how the body manages mucosal repair and inflammation in conditions like ulcerative colitis.
The study utilized a colorectal cancer cell line (DLD-1) to observe the effects of AID overexpression. Researchers identified over 600 genes induced by AID, including those within the mTOR and MAPK signaling pathways. Specifically, the expression of the NINJ2 gene increased fivefold. AID binds directly to the NINJ2 promoter and enhancer regions. Subsequently, it induces DNA demethylation, which triggers transcriptional upregulation. This molecular switch is essential for maintaining the integrity of the colonic lining.
Furthermore, the physiological impact of this pathway was tested in mouse models. AID-knockout mice showed a significant delay in recovery from induced colitis compared to wild-type mice. Because NINJ2 expression was lower in these knockout mice, the wound healing process stalled. Therefore, the researchers concluded that inflammation-driven AID upregulation is a protective response. It facilitates tissue repair by boosting NINJ2 transcription through epigenetic modifications. This process ultimately promotes faster resolution of intestinal damage.
