Researchers identified a critical mechanism by uncovering how SARS-CoV-2 nsp3 trafficking governs genomic RNA delivery. After entering the host, coronaviruses must transport their RNA to specific compartments to build replication organelles. This study demonstrates that the nsp3 protein's first transmembrane helix serves as a critical molecular address code.
The study reveals that this specific helix inserts into the endoplasmic reticulum (ER) during translation. This process relies on the host factor SEC61A1. Subsequently, the COPII factors SAR1A and LMAN1 escort the nsp3 protein to the ER-Golgi intermediate compartment (ERGIC). Consequently, this pathway ensures the viral RNA reaches the proper site for replication. Notably, researchers found that deleting key residues in this helix trapped the RNA in the cytosol. This disruption effectively crippled the virus's ability to replicate.
Moreover, the team developed a 24-residue peptide named Y24, modeled on this transmembrane helix. This peptide inhibited viral replication by interfering with the nsp3 address code. In mice, an AAV9-delivered version called A20-2 successfully reduced lung viral RNA and improved pathology scores. This host-targeted approach offers a potential broad-spectrum strategy against various coronaviruses. Because it targets conserved host pathways, it may resist the rapid mutations typical of viral proteins.
