Researchers initially investigated the Chang-Shuang-Li-Zhong powder mechanism through an integrated network pharmacology approach. Clinicians widely utilize this traditional Chinese medicine compound for managing ulcerative colitis, yet its molecular basis was previously unclear. To evaluate efficacy, researchers conducted experiments in both cellular and animal models. For instance, they activated RAW264.7 macrophages with LPS and induced colitis in mice using dextran sulfate sodium. Consequently, treatment with CSLZ significantly improved colon health by increasing length and reducing disease activity scores. In addition, the powder downregulated pro-inflammatory cytokines such as IL-1β and IL-6 at both mRNA and protein levels.
Moreover, the study identified the specific inhibition of protein phosphorylation as a primary action. Specifically, CSLZ suppresses the PI3K/Akt and MAPK pathways, including Akt, p38, ERK, and JNK. Therefore, these findings provide a scientific explanation for its clinical utility. Furthermore, this multi-target strategy positions the powder as a promising therapeutic agent for chronic intestinal inflammation. However, the researchers noted that further genetic validation is necessary to confirm these correlative findings.
