Managing patients with hereditary cancer syndromes requires precise risk data. Recent research has significantly clarified ATM variant cancer risks, providing clinicians with robust hazard ratios (HR) and cumulative risk estimates. Furthermore, this data is vital for families carrying pathogenic or predicted pathogenic variants (PV/PPV) of the ATM gene, as it informs the timing of life-saving screenings.
Specifically, researchers observed a four-fold increase in the risk of female breast cancer (HR 4.0) within a large cohort of 141 ataxia-telangiectasia families. Consequently, clinicians must prioritize specialized screening for this high-risk group. Additionally, the risk for pancreatic cancer was notably elevated, with hazard ratios of 6.6 for females and 2.8 for males. However, this study noted that these risk patterns remained similar across various family settings.
Regarding cumulative risks, the data provides a clear timeline for intervention. Moreover, for women born between 1960 and 1969, the breast cancer risk reaches 9.9% by age 50 and 40% by age 80. Notably, the lifetime risk of pancreatic cancer by age 80 is approximately 8.1% for females and 5.1% for males. This study showed no statistically significant increase in the risk of ovarian or prostate cancers, focusing clinical attention on the breast and pancreas.
The primary risks involve female breast cancer and pancreatic cancer for both genders. Female heterozygotes face a significantly higher hazard ratio compared to the general population.
