Pancreatic ductal adenocarcinoma (PDAC) represents one of the most aggressive malignancies worldwide. Consequently, its five-year survival rate remains critically low. In addition, the rising global burden of pancreatitis further complicates the landscape of pancreatic health. Both conditions involve complex stromal alterations and pro-inflammatory microenvironments. Researchers are now investigating how Vitamin C pancreatic cells interactions might offer therapeutic benefits. This nutrient possesses unique dual properties, acting as an antioxidant in inflammatory states and a cytotoxic agent in cancer.
Vitamin C primarily targets oxidative stress and inflammatory pathways in pancreatitis tissue. However, in malignant settings, it can modulate signaling pathways relevant to tumor growth. Most existing research focuses on tumor cells alone. Consequently, we lack a clear understanding of its effects on the surrounding stromal and epithelial cells. This upcoming scoping review seeks to map these interactions across both benign and malignant diseases. Specifically, it will evaluate how high-dose vitamin C influences cell-to-cell communication within the pancreatic microenvironment.
The review will follow the PRISMA-ScR guidelines to ensure methodological rigor. Furthermore, two independent reviewers will screen literature from databases like PubMed and Web of Science. They will focus on cellular-level effects, treatment durations, and various vitamin C isoforms. Therefore, the final results should provide a clear structure for future preclinical trials. By identifying knowledge gaps, this study could pave the way for more targeted therapeutic interventions.
Vitamin C can act as a pro-oxidant at high concentrations, generating hydrogen peroxide that selectively targets and destroys malignant cells while sparing healthy tissue.
The review aims to map existing literature on how Vitamin C influences the stromal and epithelial cells of the pancreas, highlighting research gaps in both cancer and inflammatory diseases.
