Researchers have achieved a breakthrough in liver-targeted mRNA delivery by developing a novel immune-modulating nanodrug. This innovation addresses a significant hurdle in genetic medicine. Currently, inflammation often hampers the success of mRNA-based therapies. Specifically, inflammatory states trigger a self-defensive response in hepatocytes. This mechanism downregulates global protein synthesis to block potential viral threats. However, it also significantly reduces the efficiency of therapeutic mRNA delivered via lipid nanoparticles (LNPs).
Furthermore, the team introduced a telodendrimer (TD) nanodrug (ND) to modulate the immune environment. They tested this technology in both human and mouse immune cells. Additionally, the researchers evaluated its effects on primary human hepatocytes (PHH). The results were impressive. The TD ND successfully inhibited endotoxin-induced inflammation. Consequently, this led to improved cell proliferation and significantly higher mRNA uptake in human monocytes and liver cells.
Moreover, the findings demonstrated that TD ND is superior to conventional steroid drugs. While steroids are common, they can sometimes impair liver recovery. In contrast, the TD ND sustained liver function in mice while rebooting the efficacy of liver-targeted mRNA delivery. Therefore, this adjuvant therapy offers a promising path for treating inflammatory liver diseases.
Inflammation acts as a biological barrier. It triggers a cellular defense that reduces mRNA expression, which prevents therapeutic genes from being translated into functional proteins.
The TD nanodrug acts as an immune modulator. It inhibits the inflammatory response that normally blocks mRNA expression, thereby enhancing the overall delivery efficiency and liver function.
