This randomized clinical trial investigated whether once-weekly semaglutide could reduce alcohol consumption and craving in adults with alcohol use disorder. Participants received low-dose semaglutide or placebo over nine weeks. Compared with placebo, semaglutide reduced the amount of alcohol consumed during a laboratory self-administration task and significantly reduced weekly alcohol craving. Some, but not all, measures of weekly alcohol consumption also improved, and participants who smoked cigarettes at baseline showed greater relative reductions in cigarettes per day. These findings provide preliminary clinical evidence that GLP-1 receptor agonists may influence reward-related behaviours beyond their established metabolic effects. However, the study was relatively small and short, so it cannot establish long-term effectiveness, relapse prevention or suitability as a routine treatment for alcohol use disorder. Larger trials are needed to clarify clinical significance, dosing and safety.

Overexpression of glycoprotein nonmetastatic melanoma protein B (GPNMB) mediates resistance to CDK4/6 inhibitors like abemaciclib and palbociclib in ER+ breast cancer through integrin-driven AKT/ERK signaling. Targeting GPNMB with antibody-drug conjugates like glembatumumab vedotin may overcome this resistance.
