Researchers have recently isolated several bioactive compounds from the fruits of Hypericum patulum, a plant commonly known as Goldencup St. John’s Wort. This discovery highlights the potential of Hypericum patulum anticancer activity in the ongoing search for natural therapeutic agents. The study identified twenty-eight prenylated acylphloroglucinols, including four novel compounds termed hypulatones C-F.
Among the newly discovered molecules, Compound 1 is particularly noteworthy as a rare meroterpenoid. It forms through the unique addition of a prenylated acylphloroglucinol unit to a sesquiterpenoid moiety. Furthermore, Compound 2, a spirocyclic polycyclic polyprenylated acylphloroglucinol, contains six chiral centers. Scientists established its relative configuration using advanced conformational analysis and NOE correlations. These structural complexities often correlate with specific biological interactions within human cells.
To assess the therapeutic potential of these isolates, the research team performed cytotoxic evaluations using the CCK-8 assay. They specifically tested the compounds against two human carcinoma cell lines: Huh-7 (hepatocellular carcinoma) and Panc-1 (pancreatic cancer). Consequently, the results revealed that compounds 6, 8, and 15 exhibited moderate antiproliferative activity. While these findings are preliminary, they provide a foundation for developing targeted natural therapies against challenging gastrointestinal malignancies.
Prenylated acylphloroglucinols (PPAPs) are a diverse class of plant secondary metabolites, primarily found in the Hypericum genus. They are known for their complex chemical structures and a wide range of biological activities, including antimicrobial and antitumor properties.
Huh-7 and Panc-1 are standard laboratory cell lines used to study liver and pancreatic cancers, respectively. Researchers use these models to screen for compounds that can effectively inhibit the growth of aggressive tumor cells.
