The CITIPARK trial evaluated citicoline 1000 mg/day as add-on therapy in Parkinson's disease. Of 485 enrolled participants, 474 were randomized (303 citicoline; 171 placebo). Citicoline significantly increased the likelihood of achieving a minimal clinically important difference in MDS-UPDRS total score (OR 2.06, 95% CI 1.06–3.99; P=0.031). MDS-UPDRS III motor scores improved by −2.97 versus −0.13 with placebo (P=0.009). CGI-II and CGI-III also improved significantly. Adverse events occurred in 22.1% vs 27.1% of participants, respectively.
