Precision oncology is rapidly evolving through the integration of artificial intelligence. Finding effective drug combinations remains a significant challenge for modern medicine. Notably, anticancer drug synergy prediction serves as a vital tool for accelerating the discovery of potent therapies. Researchers have recently developed GraFSyn, an interpretable deep learning framework designed to improve the accuracy of these predictions by focusing on chemical substructures.
Traditional molecular graph-based methods often rely on implicit feature aggregation. Unfortunately, this approach can obscure the topological identity of critical chemical components. GraFSyn addresses this limitation by using graphlet fingerprints. Specifically, these fingerprints encode drugs as explicit connected substructure units. Furthermore, the framework introduces a Dynamic Multi-Scale Convolution (DMSC) module. This module effectively learns informative representations from high-dimensional, sparse data. Consequently, the model maintains structural traceability, allowing clinicians to map signals back to specific pharmacophoric regions.
The framework also features an interaction module that captures context-dependent interactions. For instance, it evaluates the relationship between drug substructures and cell line gene expression. In general, GraFSyn demonstrated superior performance on major industry datasets. On the Merck and AstraZeneca benchmarks, the model achieved ROC-AUC values of 0.972 and 0.823, respectively. Moreover, it outperformed several representative baseline methods. Therefore, GraFSyn provides a more accurate and structurally traceable approach for anticancer drug combination screening.
