Gene duplication serves as a cornerstone of evolutionary progress. However, the role of genetic context gene duplication in determining the speed of this evolution has remained poorly understood. Consequently, a new study involving 193 mammalian genomes has shed light on how genomic location dictates the fate of new genes. Initially, researchers used microsynteny to distinguish between "source" copies in ancestral locations and "target" copies relocated to new sites. Interestingly, they found that while most paralogs evolve at similar rates, target copies often show significantly higher substitution rates. This happens because the new genomic environment provides a different regulatory context.
Specifically, the distance between copies and the size of the duplicated region influence this evolutionary speed. Furthermore, the study revealed that the selection of the fast-evolving copy often occurs in collective blocks rather than individual random choices. As a result, this block-wise polarization suggests that clusters of genes respond to their environment as a unit. Moreover, differences in cis-regulatory elements and co-expressed gene clusters between source and target copies may drive these divergent evolutionary paths. Therefore, genome architecture fundamentally shapes functional diversification over millions of years. Ultimately, these findings help us understand the precise mechanisms behind genetic variation and genome evolution. In addition, this knowledge is increasingly relevant for clinicians examining the phenotypic impacts of gene duplications and copy number variations.
