The TEMPO-2 phase 3 trial randomized 304 adults with early Parkinson's disease to flexible-dose tavapadon 5–15 mg once daily (n=151) or placebo (n=153) for 27 weeks. At week 26, the MDS-UPDRS Parts II and III combined score improved significantly more with tavapadon, with a least-squares mean decrease of 10.3 points versus 1.2 points with placebo (treatment difference −9.1; 95% CI −11.7 to −6.5; P<0.0001). Adverse events occurred in 76% versus 55%, respectively, and were mostly mild to moderate. Nausea (30% vs 3%), headache (17% vs 5%), and dizziness (16% vs 3%) were the most common adverse events.

Recent research defines ferro-aging as a conserved iron-lipid axis driving organ decline across primates. Age-related iron dyshomeostasis stimulates ACSL4-mediated lipid peroxidation, promoting cellular senescence. Notably, vitamin C directly inhibits ACSL4, suppressing phospholipid oxidation and multi-organ decay.
