These factors modify distant organs before tumor cells arrive. Therefore, clinicians can explore novel therapeutic avenues by understanding these mechanisms.
The formation of a PMN involves complex interactions between circulating elements and the microenvironment. Because factors like platelets, fibrinogen, and thrombin contribute to this preparation, they are critical targets. Platelets, for instance, release growth factors that trigger extracellular matrix remodeling. Furthermore, tissue factors activate endothelial cells. This activation increases vascular permeability. As a result, this environment allows circulating tumor cells to colonize distant tissues more effectively. Consequently, the PMN becomes a fertile ground for cancer spread.
Moreover, recognizing the role of coagulation factors in metastasis shifts our perspective on anti-thrombotic therapy. Clinical studies suggest that targeting these pathways could potentially stall cancer progression. For example, inhibiting thrombin or certain tissue factors might prevent immune cell recruitment. Additionally, this approach may reduce the overall metastatic burden in high-risk patients.
